The Holy Spirit wants to come and separate us not only from the things that harm us, but separate us to Himself and for His purposes.
1. The Holy Spirit separates you from yourself.
Now the Lord is the Spirit; and where the Spirit of the Lord is, there is liberty. But we all, with unveiled face, beholding as in a mirror the glory of the Lord, are being transformed into the same image from glory to glory, just as by the Spirit of the Lord. 2 Corinthians 3:17-18
God is transforming you more into the image of God and less into the image of who you think you are. Your sinful nature will only breed death, so the Holy Spirit wants to separate you from your old way of thinking doing what you want to do so that you can start thinking like God. Then you can receive the heart and mind of God and begin to take on his attributes of compassion, patience, holiness, etc.
The way of a fool is right in his own eyes, but he who heeds counsel is wise. Proverbs 12:15
2. The Holy Spirit separates you from the world.
As they ministered to the Lord and fasted, the Holy Spirit said, Now separate to Me Barnabas and Saul for the work to which I have called them. Then, having fasted and prayed, and laid hands on them, they sent them away. Acts 13:2-3
The Holy Spirit wants to separate you from doing what everybody else does. As Pastor Miles put it, He wants to separate you from your ghetto factor. This has nothing to do with your neighborhood or ethnicity, but rather your selfish and prideful attitude (wanting to get mine, do it my way, being all about me).
The Holy Spirit can empower you to be set free from living that way. As you are saved, you now have the ability to not retaliate when you are wronged. You are free to act differently than the world does. If you surrender yourself to the Holy Spirit, He can separate you from the worlds mentality and allow Gods mentality to reign in you.
I say then: Walk in the Spirit, and you shall not fulfill the lust of the flesh. For the flesh lusts against the Spirit, and the Spirit against the flesh; and these are contrary to one another, so that you do not do the things that you wish. Galatians 5:17
3. The Holy Spirit separates you unto Himself. Luke 4:18-19
The Holy Spirit wants you to Himself and so calls to you constantly. Your choice is to ignore Him or respond and submit to Him.

The Holy Spirit has a job for you to do according to the Lords will and plan for your life. All of these things are coordinated and strategic, but He wants your attention so that He can freely speak to you and guide you. The Holy Spirit wants you to Himself and so calls to you constantly. Your choice is to ignore Him or respond and submit to Him.
4. The Holy Spirit separates you from death. John 3:1-6
You need not fear death if Christ is our Savior, for you have the promise of new life in Him. The Holy Spirit wants to separate you from the fear of death and from death itself. He wants to give you a new life apart from your earthly one a spiritual life through which you have relationship with God.
But if the Spirit of Him who raised Jesus from the dead dwells in you, He who raised Christ from the dead will also give life to your mortal bodies through His Spirit who dwells in you. Romans 8:11
